The kill counter shown in-game is per wave, and a Totem run has 2 waves (Conjure the Totem, then Destroy it). The per-wave quota scales with both party size and difficulty: solo on Normal is 37 per wave (74 total), solo on Easy is 25 per wave (50 total), a full party of 4 on Nightmare is 180 per wave (360 total).
